PREFACE TO THE SERIES
The current volume is the ninth in the series which aims to provide comprehensive
surveys of topics in drug delivery and targeting for senior undergraduates, graduates
and those carrying out research in the many facets of the field. Drug delivery
technologies and targeting strategies grow in scope and potential, as well as complexity,
almost daily. New opportunities arise through the development of new materials for
the design and fabrication of drug delivery vehicles and carriers; new challenges
are posed by the discovery and development of new therapeutic agents, which
include not only small organic and inorganic molecules but oligomers and macro
molecules usually with no natural propensity for transport across biological barriers.
The book series has, to date, covered delivery technologies in microencapsulation1
and liposomal delivery2,3, the promotion of drug absorption4, the important issues
surrounding peptide and protein delivery5, interfacial phenomena in drug delivery
and targeting6 and biodegradable polymers of natural, synthetic or semi-synthetic
origin7. The technology of drug delivery can rarely be dealt with simply as a
materials science topic, for systems have to be evaluated in the context of the
biological environment in which they operate in vivo.
Professor Benita has edited the current volume dealing with an aspect of lipid
nanotechnology — submicron emulsions in targeting and delivery. The field evolved
from developments with intravenous fat emulsions, which are covered in Chapters
2-4 of the book. A variety of submicron systems — emulsions and lipid suspensions
(solid lipid particles) have been investigated. The next part of the book addresses
these topics. The final chapter covers perfluorochemical emulsions as blood
substitutes, an intriguing area of research. Lipidic systems have great scope once we
understand fully their physical chemistry and the manner in which they interact
with the body.
Drexler8 writes of conventional drug delivery: “Drugs are dumped into the body,
tumble and bump around in solution haphazardly until they bump a target molecule,
fit and stick ... ”. He proposed that the future of therapy will consist of nanomachines
which will gain access to diseased sites, recognize the target, disassemble damaged
parts, rebuild and reassemble. Many lipidic systems are, indeed, self-assembling. We
are perhaps still some way from that vision, but the progress that is made step by
step is the stuff of research in drug delivery and targeting. It is the role of a book
series to record and analyze that progress, and we hope that this series will achieve
those aims and provide some stimulus for further discovery.
