This workdisclosestheuseofring-openingcrossmetathesis(ROCM)of8- oxabicyclo[3.2.1]octenederivativesforthepreparationofsubstitutedpyrans,using Grubbs'ruthenium-basedmetathesiscatalyst. Theoxabicyclicsystemsweresynthesized usingFohlishconditions,whichinvolvethe[4+3]cycloadditionbetweenfuranandan oxallylcation. AnunusualinfluenceinthereactivityandselectivityoftheROCM reactionswasdiscovereduponsubstituentvariationattheC3positionoftheoxabicyclo. Theseresultsledtofurtherinvestigationsthatinvolvedthesynthesisofaseriesof8- oxabicyclo[3.2.l]octenederivativeswithdifferentsubstituentsatC3. Kinetic experimentswereconductedwiththesubstrates,usingNMRtomonitorthereactions. Theseexperimentsprovidedrelativeratesofring-openingmetathesispolymerizationfor IX theseriesofoxabicyclicderivativeswhencomparedwith4,10-dioxa-tricyclo[5.2.1.0 2'6]dec-8-ene-3,5-dione(adductoffuranandmaleicanhydride). Therelativeratetrend observedamongthesubstratesshowedthattheirreactivityisaffectedbythecombination ofelectronicandstericeffects. Theknowledgeobtainedcanbeusedtoaccelerate sluggishROCMreactionsinthesynthesisofpyransfromthesetypesofsubstrates. Substitutionatthebridgeheadpositionoftheoxabicycledecreasesthereactivityofthe systemandbringsintofocustheproblemofregioisomers. However,thecareful considerationofthesubstituentattheC3positiongavegoodyieldsofthepyrans,and excellentregioselectivitywasobtainedhavinganendo-a\coho\asasubstituentatthat position. Inaddition,theintramolecularROCMofoxabicyclicsystemhavingatethered alkeneattheC2positionwasstudiedandledtolinear-fusedpyrans. Furthermore,preliminaryworkwasdonetowardsthesynthesisoflatrunculinB usingtheROCMapproach.
