ABSTRACT
The present investigation on tomato was comprised of 13
parents (5 lines + 8 testers), their 40 F s (developed through line x tester

mating design) and two standard check hybrids (Bhagya & Abhinav).
These parents, hybrids and checks were evaluated in Randomized Block
Design with three replications. The objective of the investigation was to
generate information on magnitude of relative heterosis, heterobeltiosis
and standard heterosis, gene effects, combining ability effects of parents
and F s for quantitative and quality characters as well as elucidation of

genetic diversity among 15 tomato lines through molecular markers.
The experiment was laid out at “Research Centre for Distant
Hybridization in Field and Fruit Crops, Dept. of Agriculture
Biotechnology, Anand Agricultural University, Anand during kharif
season 2015-16. Heterosis and combining ability analysis were carried
out for twenty characters viz. days to first flowering, plant height,
number of primary branches per plant, number of fruits per cluster,
number of fruits per plant, harvest duration, fruit yield per plant, average
fruit length, average fruit girth, average fruit weight, pericarp thickness,
moisture content, total soluble solids, total soluble sugar, reducing
sugar, titratable acidity, ascorbic acid, lycopene, phenol and ToLCV
incidence.

The analysis of variance revealed that the mean square due to
genotypes, parents and hybrids were significant for all the characters
except titratable acidity for genotypes, reducing sugar and titratable
acidity for parents and pericarp thickness and titratable acidity for
hybrids, which suggested differences among themselves for all the
characters under study. The per se performance of hybrids was higher
than parents in desired directions for most of the characters suggesting
the possibility of heterotic crosses. The female parent DHT-14-8 was
identified for maximum fruit yield per plant (1261.40 g) among females
along with higher number of branches per plant (8.46) and number of
fruits per plant (87.60). The male parent EC-589496 flowered (41.60
days) earlier with highest fruit yield (1312.56 g) among male. In case of
hybrids, the highest mean value for fruit yield per plant (4160.43 g) was
depicted by DHT-14-8 x EC-589496 with higher number of fruits per
plant (640.06). Among parental genotypes, DHT-14-8, EC-589496 and
IIHR-2101 yielded superior heterotic crosses for fruit yield per plant and
its component characters.
The magnitude of heterosis for fruit yield per plant was found
positive and highly significant by the cross DHT-14-8 x EC-589496 for
relative heterosis (223.27%), heterobeltiosis (229.83%) and standard
heterosis (check 1- 545.80% & check 2- 646.13%). The analysis of
variance for both the combining ability effects was significant for all the
characters except gca variance of pericarp thickness and sca variance of
number of fruits per cluster, pericarp thickness, total soluble solids,
reducing sugar and lycopene. Prime role of additive genetic variance was
found for number of fruits per cluster, total soluble solids, reducing
sugar and lycopene.

For fruit yield per plant, among female parents, DHT-14-8,
AT-3 and ToLCV RES-3 were found good general combiners; while among
male parents, EC-589496 and IIHR-1966 were good general combiners.
The cross combinations DHT-14-8 x EC-589496, AT-3 x EC-500047 and
AT-4 x IIHR-1966 were the best for specific combining ability effects for
fruit yield per plant. The 2 2sca ratio less than unity for most of the
characters suggested preponderance of non-additive gene action.
The 15 diverse genotypes of tomato belonging to seven different
species were evaluated for genetic diversity by using PCR based
molecular markers viz. ISSR, SSR and SCAR markers. All the three
marker systems generated high levels of polymorphism. ISSR and SSR
based PCR resulted in mean PIC (Polymorphism Information content)
values of 0.79 and 0.65, respectively. The ISSR gave mean per cent
polymorphism value of 95.23%, whereas it was 100% in case of SSR.
Among types of markers, SSR gave better results as compared to
others because of huge variability present in tomato lines under study.
Total four SCAR primers gave amplification specific to resistant and
susceptible ToLCV genotypes. The banding pattern generated through
SCAR could sufficiently discriminate all the 15 genotypes for ToLCV
resistance, which also matched to field data for ToLCV incidence. Few
genotypes showed aberrant banding pattern with a specific primer due
to the presence of specific locus for ToLCV in a single genotype. These
specific SCAR amplicons might be useful for discrimination of tomato
genotypes for ToLCV resistance and molecular characterization.
Altogether the three marker systems ISSR, SSR and SCAR provided
wide genomic coverage and are, therefore, better indicators of the genetic
relationship among 15 diverse genotypes of tomato.
