SAMENVATTING
Bananen zijn één van de belangrijkste fruitgewassen die wereldwijd geproduceerd worden
en dat is eveneens het geval in Zuidoost Azië en Papoea Nieuw Guinea waar dit fruit een
stapelvoedselgewas is voor de rurale bevolking. In dit deel van de wereld worden bananen
voornamelijk geproduceerd door kleinboeren in gemengde landbouwsystemen voor de
plaatselijke markten maar in de Filippijnen vormen bananen één van de meest belangrijke
exportproducten. Zoals in andere delen van de wereld wordt ook in Azië en de Pacific de
productie van bananen belemmerd door verschillende pathogenen die een verscheidenheid
aan ziekten veroorzaken. Eén van deze ziekten wordt veroorzaakt door plantenparasitaire
nematoden waarvan Radopholus similis de belangrijkste is.
De algemene doelstelling van onze studie was om nieuwe natuurlijke bronnen van
resistentie tegen R. similis te vinden in het bananenkiemplasma van Zuidoost Azië en
Papoea Nieuw Guinea. Voor de screening experimenten werd een R. similis populatie
afkomstig uit Davao in de Filippijnen gebruikt. Aangezien deze populatie voorheen nog niet
gekarakteriseerd was was het één van onze specifieke doelstellingen om de in vitro
reproductie-capaciteit en de in vivo pathogeniciteit van deze R. similis populatie te bepalen
en te vergelijken met andere R. similis populaties. Aangezien tijdens de screening
experimenten opgemerkt werd dat verschillen in experimentele omstandigheden aanleiding
gaven tot verschillen in de bekomen resultaten was het eveneens één van onze specifieke
doelstellingen om de invloed van lokale experimentele omstandigheden op de
waardplantreactie voor R. similis van een selectie van referentie bananencultivars meer in
detail te onderzoeken.
In een eerste deel (Hoofdstuk 3) werd de in vitro reproductie-capaciteit op wortelschijfjes
en de in vivo pathogeniciteit op een aantal geselecteerde FHIA (Fundacion Hondureña de
Investigaciones Agricola) bananen hybriden van de Davao R. similis populatie bepaald. De
in vitro studie toonde aan dat deze nematodenpopulatie een korte lag-fase heeft en een hoge
maximale groeisnelheid in vergeljking met twee andere R. similis populaties afkomstig uit
de Filippijnen (Los Baños en Quezon) en twee exotische R. similis populaties afkomstig uit
Oeganda en Indonesië. De in vivo studie toonde aan dat de Davao R. similis populatie zeer
pathogeen was voor de geselecteerde FHIA bananenhybriden.

In het tweede deel (Hoofdstuk 4) werden verschillen in experimentele omstandigheden op
de waardplantreactie voor R. similis van een selectie van referentie bananencultivars nader
onderzocht. De consistentie van de waardplantreactie van de referentie bananencultivars
bekomen onder verschillende lokale experimentele omstandigheden werd met elkaar
vergeleken. Wat de densiteit van de nematodenpopulaties betreft werden significante
verschillen bekomen in zeven op de elf experimenten die uitgevoerd werden in het
nematologie laboratorium van het Bureau of Plant Industry (BPI) in Davao en in twee op de
zes experimenten die uitgevoerd werden in het nematologie laboratorium van het Institute
of Plant Breeding (IPB) in Los Baños. Hogere nematodenpopulatie densiteiten werden
bekomen wanneer de bananenplanten opgroeiden in een substraat dat meer zand bevatte. De
bekomen resultaten toonden ook aan dat de nematodenpopulatie densiteiten beïnvloed
werden door de ouderdom van de planten en de daaraan gerelateerd verschillen in
wortelontwikkeling.
In een derde deel (Hoofdstuk 5) werd de waardplantreactie (resistentie en tolerantie) van
een selectie van bananenvariëteiten afkomstig uit Papoea Nieuw Guinea en Zuidoost Azië
voor de Davao R. similis populatie geëvalueerd. Geen enkele van de onderzochte 32
variëteiten afkomstig uit Papoea Nieuw Guinea bleek resistent te zijn voor de Davao R.
similis populatie. Evenwel, vijf van de onderzochte variëteiten afkomstig uit Papoea Nieuw
Guinea vertoonden tolerantie voor R. similis infectie: Pok Pok (AAB), Ambowga (AA),
Muga (AA), Manam (AA) en Migea Arizi (AA). Drie van de onderzochte 34 variëteiten
afkomstig uit Zuidoost Azië vertoonden wel resistentie tegen de Davoa R. similis populatie:
Kluai Pa 26 (AA), K. Nang Nuan (AAB) and Pisang Papan (AAA).
In een vierde deel (Hoofdstuk 6) werd in een microplot experiment het effect van de Davao
R. similis op de groei en de opbrengst van vier populaire Aziatische bananenvariëteiten en
de vatbare referentie bananencultivar Grand Nain (AAA) onderzocht. Voor al deze
variëteiten werd een opbrengstverlies van 25 tot 68 % vastgesteld veroorzaakt door infectie
met de Davao R. similis populatie. Infectie met deze nematodenpopulatie veroorzaakte een
hogere plantenmortaliteit, “toppling” (omvallen van de planten) en verlenging van de
vegetatieve groeicyclus vergeleken met niet-geïnfecteerde planten. De laagste aantallen
nematoden werden opgemerkt in de wortels van de variëteit Latundan (AAB) en het verlies
in opbrengst was eveneens lager in deze variëteit in vergelijking met de andere onderzochte
variëteiten wat een aanduiding is dat Latundan minder vatbaar is voor R. similis.

SUMMARY

Banana is one of the most important fruits grown worldwide also in Southeast Asia and
Papua New Guinea where the fruit is used as a staple or staple substitute by the rural
population. The fruit is primarily grown by smallfarmers in mixed cropping systems and
produced for domestic markets, but in the Philippines it is one of the most important export
crops. As in other parts of the world, also in Asia and the Pacific banana is affected by
several pathogens causing a diversity of diseases. One of these diseases is caused by plant–
parasitic nematodes, especially Radopholus similis, the burrowing nematode.

It was the general objective of our study to identify new natural sources of resistance to R.
similis among the banana germplasm that originated in Southeast Asia and Papua New
Guinea. Since the Davao population of R. similis that was used for the screening
experiments had not been characterized, one of the specific objectives of our study was to
examine the reproductive fitness and pathogenicity of this nematode population. Also, since
during the screening experiments several constraints of the used protocols were observed,
another specific objective of our study was to examine the effect of the local experimental
conditions on the host response to R. similis of a selection of reference banana cultivars.

In the first part (Chapter 3), the in vitro reproductive fitness on carrot discs and the in vivo
pathogenicity on selected FHIA (Fundación Hondureña de Investigaciónes Agrícola)
hybrids of the Davao population of R. similis were investigated. It was shown that on carrot
discs this population has a short lag phase and a high maximum growth rate compared with
two other R. similis populations from the Philippines (Los Baños and Quezon) and two
exotic populations of R. similis (from Uganda and Indonesia). It was also shown that on the
selected FHIA hybrids the Davao population of R. similis is highly pathogenic.

In the second part (Chapter 4), the effect of differences in local experimental conditions on
the host response to R. similis of a selection of reference banana cultivars was investigated.
The consistency of the host response of the reference banana cultivars across the different
screening experiments that were undertaken under different local experimental conditions
was compared. For the nematode population density, significant differences between the
reference banana cultivars were observed in seven out of 11 batches carried out in the

nematology laboratory at the Bureau of Plant Industry (BPI) in Davao and in two out of six
batches carried out in the nematology laboratory at the Institute of Plant Breeding (IPB) in
Los Baños. Higher nematode population densities were observed when the banana plants
were grown in substrates with a higher proportion of sand. The results also showed that the
nematode population densities were affected by plant age and the resulting differences in
root development.

In the third part (Chapter 5), the host response (resistance and tolerance) of a selection of
Papua New Guinean and Southeast Asian banana varieties to the Davao population of R.
similis was evaluated. No sources of resistance to R. similis were found among the 32 Papua
New Guinean banana varieties evaluated. However, five varieties were identified as tolerant
to R. similis: Pok Pok (AAB), Ambowga (AA), Muga (AA), Manam (AA) and Migea Arizi
(AA). Among the 34 Southeast Asian banana varieties evaluated, three varieties were
identified as resistant to R. similis: Kluai Pa 26 (AA), K. Nang Nuan (AAB) and Pisang
Papan (AAA).

In the fourth part (Chapter 6), the yield response of four popular banana varieties from
Southeast Asia to infection with the Davao population of R. similis was investigated in a
microplot experiment. Bunch weight reduction of nematode-infected plants ranged from 25
to 68 % for all four Southeast Asian banana varieties and Grand Nain (AAA), the
susceptible reference cultivar that was also included in the experiment. Infection with the
Davao population of R. similis caused higher plant mortality, toppling and lengthening of
the vegetative growth cycle compared with uninfected plants. The banana variety Latundan
(AAB) had the lowest nematode population density and a low bunch weight reduction
compared with the other banana varieties examined which may indicate that this banana
variety is partial resistant to R. similis.
