1. Einleitung
1 Einleitung
Neoplastische Transformationen gehören mit zu den häufigsten Todesursachen
beim Hund (u.v.a.: Cotchin 1959; Dobberstein 1953; Dorn et al. 1966; Eichelberg
and Seine 1996; Eskens 1983; Gutsche 1964; MacVean et al. 1978; Priester and
Mantel 1971; Priester and McKay 1980; Überreiter 1977). Trotz gestiegener Be-
deutung molekularbiologischer Methoden (z.B. Sothern Blot, Polymerase Chain
Reaction) in der Tumordiagnostik, stellt die morphologische Bestimmung der Hi-
stogenese und der Dignität von veränderten Zellen, Zellverbänden und Geweben
weiterhin den wesentlichen und zentralen Untersuchungsschritt im Befunden und
oft daraus folgend im Prognostizieren neoplastischer Prozesse dar. Bei gut diffe-
renzierten Geschwülsten kann in der Regel mit herkömmlichen Färbetechniken
der morphologische Beziehungsnachweis zum Ursprungsgewebe geführt werden.
Für eine beträchtliche Anzahl von Tumoren ist eine solche Zuordnung zuverläs-
sig möglich. Sie bereitet jedoch in solchen Fällen Probleme, wo Tumorzellen
weitgehend entdifferenziert sind, wo aberrante oder atypische Differenzierungs-
merkmale vorliegen oder wo simultan Merkmale verschiedener Gewebetypen
auftauchen, und somit die Zuordnung nicht eindeutig festzustellen ist. Solche Tu-
moren tauchen dann in den Befunden als Blastome, großzellige, kleinzellige,
spindelzellige oder pleomorphe Neoplasien ohne weitere Spezifikationen oder
manchmal auch noch als Retikulumzellsarkome auf. Weiterhin treten Zuord-
nungsprobleme auf, wenn Metastasen ohne Primärtumor gefunden werden. In
solchen Fällen kann versucht werden, mit Hilfe von Enzymhistochemieund/ oder
Elektronenmikroskopie nach gewebespezifischen Strukturen oder Gewebepro-
dukten zu suchen. Gerade bei Tumoren mit niedrigem Differenzierungsgrad miß-

toplasmatische Differenzierungsprodukte (zum Beispiel: Enzymmuster, Verhor-
nung von Plattenepithel oder Sekretion von Drüsen) desorganisiert werden.
In den letzten Jahren konnte in erster Linie an Geweben des Menschen gezeigt
werden, daß spezifische Strukturen des Zytoskeletts auch bei weitgehender Ent-
differenzierung weiterhin nachweisbar bleiben (Osborn and Weber 1983; Quinlan
et al. 1985). Dabei hat die Weiterentwicklung der Immunhistochemie, insbeson-
dere seit Einführung der monoklonalen Antikörpertechnik (Köhler and Milstein
1975) in Verbindung mit Differenzierungs- bzw. Funktionsmarkern des Zytoske-
letts, der morphologischen Diagnostik neue Anstöße gegeben, diesem Dilemma
zu entkommen. Der große Vorteil dieser Methoden besteht darin, daß spezifische
Marker auf der Basis der lichtmikroskopischen Morphologie in situ nachgewie-
