THE UKNCC
Introduction
The United Kingdom National Culture Collection (UKNCC) co-ordinates some of the marketing and
research activities of the UK’s public domain service culture collections, promoting collaboration between
the different centres. The UKNCC Member Collection holdings number more than 73000, made up of c.
2300 algae and protozoa, 20000 animal cell lines, 25000 bacteria, 25000 fungi including yeasts, plus
actinomycetes, cyanobacteria, nematodes, mycoplasma and viruses. The supply of these organisms,
associated services and the expertise of the nine national culture collections that make up the UKNCC are
available to support your microbiological and cell culture needs. Visit the website to see what is available

generally in identification and preservation but they, and their parental organisations, offer many more
services.

This catalogue lists the algae (including cyanobacteria) and protozoa available from the UKNCC Member
Collections and presents information on their isolation, the host or substrate from which they were isolated
and how they are preserved/maintained. Further information can be accessed at the UKNCC website or
directly from the Collection or its website. Not all the holdings of the UKNCC are listed in the catalogues or
the publicly available database; several thousand strains are held in reserve because they are incompletely
named, or for other reasons. If you are interested in access to such strains contact the relevant UKNCC
Collection direct for the organism type you require. If you wish to receive specialist lists of organisms based

UKNCC Quality Policy
It is the policy of the UKNCC to endeavour to provide its clients on every occasion with the products and
services that they require. The products and services supplied are of marketable quality and will fulfil
product claims as defined in the individual member collection catalogues or website. At all times good
techniques and procedures, as defined in the individual collection’s laboratory procedures manual or
standard operating procedures, will be in operation and regular audits carried out to ensure that these
procedures are followed and are effective.
The UKNCC Collections either employ a recognised accreditation scheme e.g. ISO 9000 or follow the
UKNCC Quality Management System. The UKNCC Quality Manual is available on the UKNCC website:

Convention on Biological Diversity (CBD)
The UK is a full signatory to the Convention on Biological Diversity (CBD) and consequently UK national
collections must receive and distribute strains within the spirit of the CBD. All legislation and policy on
intellectual property rights governing actions of the UKNCC Member Collections will be followed. Each
party must make reasonable efforts to ensure the depositor has permission from the appropriate authority to
deposit the material in the collection, and that conditions for further distribution are defined in a manner
compliant with the UKNCC stance on the CBD.

UKNCC MEMBER COLLECTIONS
There are nine public service Member Collections of the UKNCC with one (CCAP) split on two sites.
These are listed below and together provide a comprehensive coverage of microorganisms and cell-lines,
including algae, animal cell-lines, bacteria, bacteriophage, cyanobacteria, fungi (including yeasts),
mycoplasma, nematodes, plasmids, protozoa, transposons and viruses. Hard copy, electronic and on-line
catalogues are available listing all these strains. The Collections operate to quality standards and do not only
supply organisms, but also provide several other related services.

Culture Collection of Algae and Protozoa Oban and Windermere sites
The Culture Collection of Algae and Protozoa (CCAP) is the oldest of the major protistan service culture
collections. It originated, in the 1920's, from the research collection of Prof. E.G. Pringsheim at the German
University of Prague. In the following years the Collection has developed and expanded. The CCAP was
briefly located in London and then in Cambridge from the date of wartime evacuation in the early 1940's to
1986. The CCAP has operated at two sites since 1986. Protozoa and freshwater algae are maintained at the
Natural Environment Research Council (NERC), Centre for Ecology & Hydrology Windermere. Marine
algae are maintained at the NERC Dunstaffnage Marine Laboratory, Oban. The Collection maintains a wide
range of microalgal taxa, including cyanobacteria, small thalloid red algae and free-living non-pathogenic
protozoa. The CCAP is an International Depository Authority (IDA) for the groups of organisms described
above, under the terms of the Budapest Treaty (1977).

Equipment/Techniques
• Fully equipped purpose-built laboratory suites, including separate, dedicated, media-prep and wash-up
facilities

• Various small incubators both illuminated and non-illuminated

• Cryopreservation facilities allowing controlled-rate, uncontrolled two-step cooling and vitrification

• Molecular biology equipment: PCR machine, gel electrophoresis apparatus, DNA sequencing apparatus,
pulse-field gel electrophoresis apparatus
Core research
• Cryopreservation: protocol development and improvement; mechanisms of lethal and sub-lethal
cryoinjury.
• Taxonomic: using both traditional and modern molecular techniques.
Contract research
Wide range of areas and techniques including biochemical, biotechnological, ecological and microbiological
approaches have been used to fulfil contract research projects including discrete CCAP projects and input
into larger CEH projects.

The Culture Collection of Algae and Protozoa (Oban)
The CCAP Marine Collection comprises a wide range of marine cyanobacteria and marine and hypersaline
taxa from most algal classes. The strains represent material collected from a wide range of geographical
locations and ecological niches, ranging from Antarctic seas to soda lakes in Africa. The Collection also
maintains a range of small multicellular seaweeds. Most material is held as live cultures maintained by
regular serial subculturing, however, some material is cryopreserved. The CCAP is an IDA for patented
strains and can accept valuable strains for confidential safekeeping. The Collection, with its associated
facilities for cultivating algae under controlled conditions, provides an ideal environment for experimental
and medium (pilot) scale algal culture work.
Equipment/Techniques
CCAP marine facilities at Oban comprise a fully equipped algal culture and microscopy laboratory with
laminar flow cabinets and basic cryopreservation facilities, and several microscopes, of which one is
equipped with bright field, phase/interference contrast, epi-fluorescence and digital imaging. The adjoining
media-prep/washing facility houses two autoclaves and a distilled water supply. Two walk-in constant
temperature (10-30°C) rooms and facilities for larger scale (200-300 litre in total) algal cultivation, and
seven constant temperature, illuminated algal culture cabinets, are available for culturing at temperatures
ranging from 0°C to 40°C (±1°C). Facilities are also available for culturing under modified atmospheres
(e.g. increased CO ). The CCAP molecular genetics facility is equipped to carry out molecular biological

studies, DNA sequencing and the development and screening of a variety of molecular markers. Equipment
includes PCR machines, gel electrophoresis equipment, DNA extraction and purification, electrophoresis
documentation and digital imaging darkroom. The Collection and associated staff undertakes core research
on algal culture, algal phylogeny and systematics, toxic algal biology and ecology, and algal molecular
genetics. Contract research in these and related areas can be accommodated by arrangement with CCAP
staff.
Additional facilities of DML
The general facilities of DML available to CCAP include TEM, SEM, centrifuges, HPLC, GC, Fluorometer,
AA photospectrometer, X-ray Fluorescence Spectrometer, Inductively Coupled Plasma Mass Spectrometer
(ICP-MS), scintillation counter, autoanalyser, and a dedicated radiation suite. There are seagoing boats and
extensive experimental aquarium facilities supplied with running seawater. The Collection also has access to
DML computing and Internet facilities, and an excellent library collection of journals and books.

The CCAP provides a number of algal strains that are routinely used in ecotoxicity testing. These can
normally be supplied at 2 weeks’ notice.
