Abstract Iron is the most abundant trace element in the human body. It is well
knownthatironisanimportantcomponentofhemoglobininvolvedinthetransport
ofoxygen.Asacomponentofvariousenzymes,itparticipatesinthetricarboxylic
acidcycleandoxidativephosphorylation.Ironinthenervoussystemisalsoinvolved
inthemetabolismofcatecholamineneurotransmittersandisinvolvedintheforma-
tionofmyelin.Therefore,ironmetabolismneedstobestrictlyregulated.Previous
studieshaveshownthatirondeficiencyinthebrainduringinfantsandyoungchil-
drencausesmentalretardation,suchasdelayeddevelopmentoflanguageandbody
balance, and psychomotor disorders. However, if the iron is excessively deposited
intheagedbrain,itiscloselyrelatedtotheoccurrenceofvariousneurodegenerative
diseases,suchasAlzheimer’sdisease,Parkinson’sdisease,andFriedreich’sataxia.
Therefore,itisimportanttofullystudyandunderstandthemechanismofbrainiron
metabolismanditsregulation.Onthisbasis,exploringtherelationshipbetweenbrain
ironregulationandtheoccurrenceofnervoussystemdiseasesanddiscoveringnew
therapeutictargetsrelatedtoironmetabolismhaveimportantsignificanceforbreak-
ingthroughthelimitationofpreventionandtreatmentofnervoussystemdiseases.
Thisreviewdiscussesthecompleteresearchhistoryofironanditssignificantrolein
thepathogenesisofthecentralnervoussystem(CNS)diseases.
