A.1 Definition of side effect frequency
Common: Happens to 1 out of 10 patients
Occasional: Happens to > 1 out of 100 patients and < 1 out of 10 patients
Rare: Happens to > 1 out of 1000 patients and < 1 out of 100 patients
Extremely rare: Happens to 1 out of every 10,000 patients, and single case reports
Immediate: Within 1-2 days
Prompt: Within 2-3 weeks
Delayed: Any time later during therapy
Late: Any time after completion of treatment
A.1.1 Actinomycin D, Dactinomycin

Mechanism of action: Cell cycle phase non-specific. Inhibition of DNA-primed RNA synthesis by
intercalating with guanine residues of DNA. At higher concentrations, inhibition of DNA synthesis,
interstrand and DNA-protein crosslinks
Application: intravenously
Metabolism: Actinomycin D is partially metabolised (15%) in the liver into inactive metabolites.
15% intact drug are excreted in the faeces and 15 % intact drug are excreted within one week
after application in the urine.
Terminal half-life: 24-48 hours.
Contraindication: Hypersensitivity to actinomycin D
