Introduction 

Dendritic cells and innate immunity 

Dendritic cells (DCs) have been recognized as central players in immune regulation since their 
discovery by  Ralph Steinman and Zanvil Cohn in the 1970s. 1–3 DCs sense pathogen stimuli 
​
through pathogen-recognition receptors (PRRs) and decode, process, and present information to 
lymphocytes to stimulate  or suppress immune responses. To regulate the quality of immune 
responses, several functionally specialized subsets of DCs exist, but it has become evident that 
DCs display considerable plasticity to integrate different stimuli and elaborate more complex and 
variable responses. 

The first records available that speculated about how the immune system works came from Frank 
Burnet 4 and David Talmage. 5  They hypothesized that an enormously diverse repertoire of 
​ ​
lymphocyte  antigen-binding  receptors  allows  the  immune  system  to  recognize  virtually  any 
molecule  with  high  specificity.  The  link  between  lymphocyte  antigen  recognition  and  the 
well-known capacity of engulfment of pathogens by phagocytes 6 only became evident when Nossal 
​
and colleagues 7 identified the accumulation of radiolabeled antigens in reticular cells in the B cell 
​
follicles of murine lymph nodes and spleens, with a consequent increase in antibody titers. These

cells, later found to be DCs and integrated in the mononuclear phagocyte system by van Furth, 8 
​
were shown to be the key initiators and regulators of immune responses. Charles Janeway Jr., in 
1989, hypothesized that DCs can recognize conserved features of microbial pathogens (also known 
as pathogen-associated  molecular  patterns, PAMPs) through PRRs. This mode of recognition 
induces inflammatory responses, innate host defenses and adaptive immunity.  

Steinman’s seminal research on the contribution of PRRs to innate immune was complemented by 
studies showing how DCs resort to ancient stress response pathways, such as mechanisms that 
involve amino acid and endoplasmic reticulum stress control, to program immune responses and 
properly stimulate lymphocytes. 9–11 
​

DCs  develop  in  the  bone  marrow  from  hematopoietic  stem  cells  that differentiate  into DC 
precursors. When DC precursors migrate to tissues via the bloodstream, they become immature 
DCs, which are characterized by low levels of Class II MHC products (MHCII) and ability to 
capture pathogens through both non-receptor-mediated mechanisms and PRRs. 12–14 Immature 
​
DCs can vary abundantly in respect to their expression of PRRs, location, microenvironment, 
cytokine  secretion  and  type  of  immune  response  triggered.  12–14​15  These  differences  in  DC 
​
phenotype have been used to categorize distinct DC subtypes. 13,16 It is important to stress that 
​
despite the superior capacity of DCs to stimulate T cells, they may orchestrate innate immune 
responses independently of T cell activation or migration to secondary lymphoid organs. 17,18 
​

Subtypes of dendritic cells 

Murine DCs can be divided into classical (cDCs) and plasmacytoid DCs (pDCs). Despite their 
similar origin, pDCs and cDCs have different life cycles. While pDCs terminally differentiate in 
the bone marrow, 19 the precursors of cDCs migrate to lymphoid and non-lymphoid organs, where 
​
they terminally differentiate into the different subsets. 20,21 cDCs are identified by their higher 
​
levels of CD11c and Class II MHC, and are classified depending on their levels of CD8α or 
CD11b, 22,23​ as well as Th polarization capacity 24–27​ (Table 1).
